Accessing EVs and characterizing them in vivo in relation to bone remodeling processes presents major technical challenges due to the size of some EVs and their location at the interface between mineralized and non-mineralized tissues. The VINCI project addresses the global characterization challenge of (sub)nanoscale chemistry, three-dimensional structure and chemical coordination of biological structures at solid-liquid interfaces, by developing new, damage-limiting electron microscopy characterization methods. More specifically, this project aims to develop new electron microscopy methods to identify and characterize at high spatial resolution the extracellular vesicles produced in beam-sensitive bone, which are essential for understanding bone remodeling and repair processes. More broadly, the new characterization methods and protocols developed as part of this project will open up new perspectives on the study of biomaterial-cell interfaces, the relevant changes underlying bone pathology or the study of interfaces between soft and hard materials, not only for the life sciences but also for a number of material science challenges.
